Analysis of ecdysteroid conjugates: Chromatographic characterization of sulfates, phosphates and glucosides

Ecdysone monosulfates (at C-2, C-3 and C-22) and ecdysone 2,22-disulfate were synthesised using sulfur trioxide triethylamine complex as sulfation agent. Direct sulfation of ecdysone afforded a mixture of various sulfates. Regioselective sulfation of ecdysone was achieved by utilizing temporary protection of hydroxy groups. The chromatographic behaviour of synthetic sulfates was compared with that of various other polar conjugates of ecdysone and 20-hydroxyecdysone. General rules to characterise ecdysteroid conjugates are proposed.
